Your ear gets a pain when a plane takes off, due to the difference of the pressure on the two sides of the eardrum. At the ground level, the pressure is the same on either sides of your eardrum so it does not cause a pain. But when you ascend in an airplane, the air pressure decreases, while the air trapped in your inner ear will cause your eardrums to push outward.Due to this, you will feel a discomfort and also a pain in your ears.
